RESUMO

Canada has been a pioneer in adopting a harm reduction approach to address risks associated with drug use for people who inject drugs. Today, Canada is home to 39 supervised injection sites spread throughout the country. The scientific literature demonstrates, unequivocally, that these sites have numerous health benefits for people who inject drugs, namely by decreasing risks of blood-borne diseases, overdose, and mortality. Yet, a lack of clear guidelines on optimal locations for the implementation of such sites and NIMBYISM ("Not In My Back Yard") have been stumbling blocks for planned and operating sites. Various Canadian governments have introduced their own policies to overcome the lack of national public health guidelines on community planning. Namely, policies aim to limit the exposure to sites and drug use for vulnerable populations, such as children. However, there is a veritable lack of research on the public health impacts of supervised injection sites for local communities, who tend to be disadvantaged. The existing literature fails to address the broader and differential impacts of such sites for local vulnerable and disadvantaged populations, including use of active transportation, psychological distress, perceived safety, and social cohesion. Moreover, existing research, largely focusing on assessing pre-implementation social acceptability and post-implementation impacts on crime, faces important methodological limitations. The following commentary reviews the existing literature and makes recommendations for future public health research on the impacts of supervised injection sites.


RéSUMé: Le Canada a été un pionnier dans l'adoption de l'approche de réduction des méfaits liés à la consommation de drogues pour les personnes utilisatrices de drogues par injection. Aujourd'hui, le Canada compte 39 sites d'injection supervisée répartis sur l'ensemble du territoire. La littérature scientifique démontre sans équivoque que ces sites présentent de nombreux avantages pour la santé des personnes utilisatrices de drogues par injection, notamment en réduisant les risques de maladies transmises par le sang, de surdoses et de décès. Toutefois, l'absence de lignes directrices claires sur les emplacements optimaux pour l'implantation de ces sites et le phénomène du « pas dans ma cour ¼ ("Not In My Back Yard") représentent des défis pour les sites existants et futurs. Plusieurs gouvernements canadiens ont introduit des politiques pour pallier l'absence de lignes directrices nationales en matière de santé publique et de planification communautaire. Ces politiques visent à limiter l'exposition aux sites d'injection supervisée et à la consommation de drogues pour les populations vulnérables, comme les enfants. Cependant, il y a un véritable manque de connaissances quant aux impacts en matière de santé publique des sites d'injection supervisée pour les communautés locales, qui ont tendance à être défavorisées. La littérature existante n'aborde pas les impacts élargis et différentiels de ces sites pour les populations vulnérables et défavorisées résidant à proximité, particulièrement sur l'utilisation des transports actifs, la détresse psychologique, la perception de sécurité et la cohésion sociale. En outre, la recherche existante, qui se concentre principalement sur l'évaluation de l'acceptabilité sociale avant l'implantation des sites et sur les impacts sur la criminalité après l'implantation des sites, se heurte à d'importantes limites méthodologiques. Ce commentaire fait état de la littérature existante et formule des recommandations pour de futures recherches sur les impacts en matière de santé publique des sites d'injection supervisée.

Long noncoding RNAs (lncRNAs) are major components of cellular transcripts that are emerging as important players in various biological pathways. Due to their specific expression and functional diversity in a variety of cancers, lncRNAs have promising applications in cancer diagnosis, prognosis, and therapy. Studies have shown that lncRNA DiGeorge syndrome critical region gene 5 (DGCR5) with high specificity and accuracy has the potential to become biomarkers in cancers. LncRNA DGCR5 can be noninvasively extracted from body fluids, tissues, and cells, and can be used as independent or auxiliary biomarkers to improve the accuracy of diagnosis or prognosis. Now, the underlying mechanisms of lncRNAs such as DGCR5 were explored as therapeutic targets, which have been investigated in clinical trials of several cancers. The DGCR5 lacks an appropriate animal model, which is necessary to gain greater knowledge of their functions. While some studies on the uses of DGCR5 have been carried out, the small sample size makes them unreliable. In this review, we presented a compilation of recent publications addressing the potential of lncRNA DGCR5 that could be considered as biomarkers or therapeutic targets, with the hopes of providing promised implications for future cancer therapy.

Immunometabolism, which studies cellular metabolism and immune cell function, is a possible cancer treatment. Metabolic pathways regulate immune cell activation, differentiation, and effector functions, crucial to tumor identification and elimination. Immune evasion and tumor growth can result from tumor microenvironment metabolic dysregulation. These metabolic pathways can boost antitumor immunity. This overview discusses immune cell metabolism, including glycolysis, oxidative phosphorylation, amino acid, and lipid metabolism. Amino acid and lipid metabolic manipulations may improve immune cell activity and antitumor immunity. Combination therapy using immunometabolism-based strategies may enhance therapeutic efficacy. The complexity of the metabolic network, biomarker development, challenges, and future approaches are all covered, along with a summary of case studies demonstrating the effectiveness of immunometabolism-based therapy. Metabolomics, stable isotope tracing, single-cell analysis, and computational modeling are also reviewed for immunometabolism research. Personalized and combination treatments are considered. This review adds to immunometabolism expertise and sheds light on metabolic treatments' ability to boost cancer treatment immunological response. Also, in this review, we discussed the immune response in cancer treatment and altering metabolic pathways to increase the immune response against malignancies.

Cyclooxygenase (COX) enzymes are pivotal in inflammation and cancer development. COX-2, in particular, has been implicated in tumor growth, angiogenesis, and immune evasion. Recently, COX-2 inhibitors have arisen as potential therapeutic agents in cancer treatment. In addition, combining COX inhibitors with other treatment modalities has demonstrated the potential to improve therapeutic efficacy. This review aims to investigate the effects of COX inhibition, both alone and in combination with other methods, on signaling pathways and carcinogenesis in various cancers. In this study, a literature search of all major academic databases was conducted (PubMed, Scholar google), including the leading research on the mechanisms of COX-2, COX-2 inhibitors, monotherapy with COX-2 inhibitors, and combining COX-2-inhibitors with chemotherapeutic agents in tumors. The study encompasses preclinical and clinical evidence, highlighting the positive findings and the potential implications for clinical practice. According to preclinical studies, multiple signaling pathways implicated in tumor cell proliferation, survival, invasion, and metastasis can be suppressed by inhibiting COX. In addition, combining COX inhibitors with chemotherapy drugs, targeted therapies, immunotherapies, and miRNA-based approaches has enhanced anti-tumor activity. These results suggest that combination therapy has the potential to overcome resistance mechanisms and improve treatment outcomes. However, caution must be exercised when selecting and administering combination regimens. Not all combinations of COX-2 inhibitors with other drugs result in synergistic effects; some may even have unfavorable interactions. Therefore, personalized approaches that consider the specific characteristics of the cancer and the medications involved are crucial for optimizing therapeutic strategies. In conclusion, as monotherapy or combined with other methods, COX inhibition bears promise in modulating signaling pathways and inhibiting carcinogenesis in various cancers. Additional studies and well-designed clinical trials are required to completely elucidate the efficacy of COX inhibition and combination therapy in enhancing cancer treatment outcomes. This narrative review study provides a detailed summary of COX-2 monotherapy and combination targeted therapy in cancer treatment.

N-acetyltransferase 10 (NAT10), a versatile enzyme, has gained considerable attention as a significant player in the complex realm of cancer biology. Its enigmatic role in tumorigenesis extends across a wide array of cellular processes, impacting cell growth, differentiation, survival, and genomic stability. Within the intricate network of oncogenic signaling, NAT10 emerges as a crucial agent in multiple cancer types, such as breast, lung, colorectal, and leukemia. This compelling research addresses the intricate complexity of the mechanistic role of NAT10 in cancer development. By elucidating its active participation in essential physiological processes, we investigate the regulatory role of NAT10 in cell cycle checkpoints, coordination of chromatin remodeling, and detailed modulation of the delicate balance between apoptosis and cell survival. Perturbations in NAT10 expression and function have been linked to oncogenesis, metastasis, and drug resistance in a variety of cancer types. Furthermore, the bewildering interactions between NAT10 and key oncogenic factors, such as p53 and c-Myc, are deciphered, providing profound insights into the molecular underpinnings of cancer pathogenesis. Equally intriguing, the paradoxical role of NAT10 as a potential tumor suppressor or oncogene is influenced by context-dependent factors and the cellular microenvironment. This study explores the fascinating interplay of genetic changes, epigenetic changes, and post-translational modifications that shape the dual character of NAT10, revealing the delicate balance between cancer initiation and suppression. Taken together, this overview delves deeply into the enigmatic role of NAT10 in cancer, elucidating its multifaceted roles and its complex interplay with oncogenic networks.

Since cancer is one of the world's top causes of death, early diagnosis is critical to improving patient outcomes. Artificial intelligence (AI) has become a viable technique for cancer diagnosis by using machine learning algorithms to examine large volumes of data for accurate and efficient diagnosis. AI has the potential to alter the way cancer is detected fundamentally. Still, it has several disadvantages, such as requiring a large amount of data, technological limitations, and ethical concerns. This overview looks at the possibilities and restrictions of AI in cancer detection, as well as current applications and possible future developments. We can better understand how to use AI to improve patient outcomes and reduce cancer mortality rates by looking at its potential for cancer detection.

Due to the growing demand for detection technologies, there has been significant interest in the development of integrated dual-modal sensing technologies, which involve combining two signal transduction channels into a single technique, particularly in the context of food safety. The integration of two detection signals not only improves diagnostic performance by reducing assumptions, but also enhances diagnostic functions with increased application flexibility, improved accuracy, and a wider detection linear range. The top two output signals for emerging dual-modal probes are fluorescent and colorimetric, due to their exceptional advantages for real-time sensitive sensing and point-of-care applications. With the rapid progress of nanotechnology and material chemistry, the integrated colorimetric/fluorimetric dual-mode systems show immense potential in sensing foodborne pathogenic bacteria. In this comprehensive review, we present a detailed summary of various colorimetric and fluorimetric dual-modal sensing methods, with a focus on their application in detecting foodborne bacteria. We thoroughly examine the sensing methodologies and the underlying principles of the signal transduction systems, and also discuss the challenges and future prospects for advancing research in this field.

Background: Intimate partner violence (IPV) affects many individuals and can have a significant impact on their health and well-being. In order to inform prevention strategies, several studies have focused on the determinants of IPV. However, knowledge on the association between neighbourhood characteristics and IPV remains scarce. The social disorganization theory posits that certain neighbourhood characteristics are associated with violent behaviours. This theory has been used to explain spatial variations in IPV, but most studies have been conducted in the United States. Little is known about the effect of neighbourhood factors in urban contexts outside of the United States. Design and methods: Using police data from 2016 and 2017, this study estimated the association between sociodemographic characteristics of neighbourhoods (socioeconomic status, single-parenthood, residential instability and ethnocultural heterogeneity) and IPV victimization in Montréal, Québec. Results: Results suggest a neighbourhood-level variation in IPV, and that neighbourhood-level characteristics are associated with IPV victimization. Specifically, the likelihood of IPV is higher in neighbourhoods with the lowest SES level (OR = 2.80, 95%CI: 2.47-3.17, p < 0.001) and the lowest level of residential instability (OR = 0.81, 95%CI: 0.70-0.93, p = 0.003) as well as the highest proportion of single-parent households (OR = 1.88, 95%CI: 1.65-2.15, p < 0.001). Conclusion: Although neighbourhood-level interventions to reduce IPV are rare, our results highlight the importance of developing such preventive strategies. Prevention programs targeting high-risk neighbourhoods may prove effective in reducing IPV.

Many adolescents experience violence in the context of dating and romantic relationships. Neighborhoods can influence dating violence by offering certain resources which can provide social support and opportunities for social participation, but knowledge about these effects is still limited. The purpose of the current study was to (a) assess the association between neighborhood social support, social participation, and dating violence, and (b) explore possible gender difference in these associations. This study was conducted on a subsample of 511 participants living in Montréal from the Québec Health Survey of High School Students (QHSHSS 2016-2017). QHSHSS data were used to measure psychological and physical/sexual violence (perpetration and victimization), neighborhood social support, and social participation, as well as individual and family covariates. Several neighborhood-level data from multiple sources were also used as covariates. Logistic regressions were performed to estimate associations between neighborhood social support and social participation, and Dating violence (DV). Analyses were conducted separately for girls and boys to explore possible gender differences. Findings suggest that girls who reported high neighborhood social support had a lower risk of perpetrating psychological DV. High social participation was associated with a lower risk of perpetrating physical/sexual DV for girls, whereas it was associated with a higher risk of perpetrating psychological DV for boys. Preventive strategies to foster social support in neighborhoods, such as mentoring programs, and the development of community organizations to increase the social participation of adolescents could help reduce DV. To address the perpetration of DV by boys, prevention programs in community and sports organizations targeting male peer groups should also be developed to prevent these behaviors.

BACKGROUND: Dating violence (DV) is a public health problem that could have serious repercussions for the health and well-being of a large number of adolescents. Several neighborhood characteristics could influence these behaviors, but knowledge on such influences is still limited. This study aims at (1) evaluating the associations between neighborhood characteristics and DV, and (2) assessing how spatial scale influences the estimations of the latter associations. METHODS: The Québec Health Survey of High School Students (2016-2017) was used to describe DV. Neighborhoods were operationalized with polygon-based network buffers of varying sizes (ranging from 250 to 1000 m). Multiple data sources were used to describe neighborhood characteristics: crime rate, alcohol outlet density (on-premises and off-premises), walkability, greenness, green spaces density, and youth organizations density. Gendered-stratified logistic regressions were used for assessing the association between neighborhood characteristics and DV. RESULTS: For boys, off-premises alcohol outlet density (500 m) is associated with an increase in perpetrating psychological DV. Crime rate (500 m) is positively associated with physical or sexual DV perpetration, and crime rate (250 m) is positively associated with physical or sexual DV victimization. Greenness (1000 m) has a protective effect on psychological DV victimization. For girls, walkability (500 m to 1000 m) is associated with a decrease in perpetrating and experiencing psychological DV, and walkability (250 m) is negatively associated with physical or sexual DV victimization. CONCLUSIONS: Several neighborhood characteristics are likely to influence DV, and their effects depend on the form of DV, gender, and spatial scale. Public policies should develop neighborhood-level interventions by improving neighborhood living conditions.

ABSTRACT In today's web world, Service-oriented architectures represent the main standard for IT infrastructures. Certainly, with the initiation of service oriented architecture, Web services have gained incredible growth. Web service discovery has become increasingly more significant as the existing use of web service. Discovering most appropriate web service from vast collection of web services is very decisive for successful execution of applications. In automation of web service discovery, there is always a need to deem Quality of Service (QoS) attributes during matching. A study of literature concerning the evolution of different web service discovery optimization methods with unique prominence to quality motivated service discovery have been carried out in this work. This paper depicts Bio-inspired algorithms optimizing the discovery process for semantic web services. Bio-inspired algorithm is a metaheuristics method that mimics the nature in order to unravel optimization difficulty and evaluates the analysis of some popular bio-inspired optimization algorithm systematically. This paper also focused on the principle of each algorithm and their application with respect to run time oriented QoSattributes and from result the best suitable bio-inspired optimization algorithm is been deployed.
